Bovine collagen type VI (placenta), pepsin-solubilised, 1 mg
Type VI collagen is the microfibrillar, beaded-filament collagen that anchors cells and basement membranes to the interstitial matrix and is deficient in Bethlem myopathy and Ullrich muscular dystrophy. This preparation is purified from bovine placenta by pepsin solubilisation and salt fractionation (≥90%) and lyophilised salt-free, 1 mg. Use it as a type VI standard and control antigen in ELISA, Western blotting and immunoprecipitation, or as a substrate in cell-adhesion assays. Note the slow dissolution described below.
Source
Bovine placenta
Purity
Bovine collagen type VI 90%
Format
Lyophilized, salt-free
Purification
Collagen was extracted from washed dissected tissue into dilute acetic acid after pepsin treatment. Collagen type VI was purified by using differential salt precipitation
Applications
ELISA, Immunoprecipitation, Western Blotting
Reconstitution
Collagen type VI dissolves very slowly in 0.5 M acetic acid and requires both vigorous mixing and time.
Add small amount of 0.5 M acetic acid, pH 2.5 and use Potter homogenizer (or similar) to break up the protein cake into suspension. Add 0.5 M acetic acid, pH 2.5 to yield 0.1 mg/ml concentration and use vigorous stirring for several days.
Concentration higher than 0.1 mg/ml in acetic acid requires even longer time. Use 6M urea (or other chaotropic agent) if application allows it.
Storage
Lyophilized protein: shipping at ambient temperature, long term storage (2 years) at -20°C or lower. Storage of reconstituted protein 1 month at +4°C
