Human apolipoprotein B-100 (ApoB), delipidated from plasma LDL, 0.5 mg
Native apolipoprotein B-100, the 550 kDa structural protein of low-density lipoprotein and ligand of the LDL receptor, isolated from virus-screened human plasma by density-gradient ultracentrifugation, delipidation and gel filtration to at least 90 % purity. Supplied as 0.5 mg in a carbonate/deoxycholate solution. Used as an ApoB calibrator, immunogen (1) and ELISA antigen in lipoprotein and atherosclerosis research; matching antibodies are 447 and 843.
Source
Human plasma. Donors tested negative for HBsAg, HBc, HIV-I, HIV-II and HCV antibodies
Purity
At least 90% pure, judged by SDS-PAGE. Band at 550 kDa
Format
Solution containing 50 mM Na2CO3, pH 10, 50 mM NaCl and deoxycholate. Non-sterile
Purification
Ultracentrifugation in density gradient. Delipidation with diethyl ether and ethanol. Gel filtration on Sephacryl S-300
Applications
Apolipoprotein B standard. Antigen for immunisation (1) antibody production. ELISA
