Mouse anti-human CD61 (integrin β3 / platelet GPIIIa) monoclonal antibody, clone CRC54, 1 ml
Mouse IgG1κ monoclonal against CD61, the β3 integrin subunit shared by the platelet fibrinogen receptor GPIIb/IIIa (αIIbβ3) and the vitronectin receptor αVβ3. CRC54 binds an N-terminal epitope of GPIIIa and reacts preferentially with the ligand-occupied, activated conformation (a LIBS-type epitope), which makes it useful not only for identifying platelets, megakaryocytes and megakaryoblasts by flow cytometry, immunofluorescence and IHC on frozen sections, but also for detecting platelet activation and for studying Glanzmann thrombasthenia (no reaction with GPIIIa-deficient platelets). Purified, lyophilised; reconstitute with 1 ml water.

Specificity

Reacts with platelet glycoprotein gpIIIa/VNR beta chain. This glycoprotein is the common beta chain (beta3 Integrin) of the cytoadhesins, gpIIbIIIa and VNR. Reacts with platelets and megakaryocytes. The antibody reacts preferentially with the activated/occupied form of platelet GPIIb-IIIa (ligand-induced binding site or LIBS epitope) and dissociated form of GPIIIa. CRC54-epitope is located within the N-terminal region of GPIIIa (first 100 amino acids). CRC54 does not react with platelets of patients with Glanzmann thrombastenia
